JotDyes – Extracellular vesicles membrane labeling dyes
The world’s first membrane labeling dyes that span the full lipid bilayer. These intelligent dyes only ‘light up’ after embedding into the lipid bilayer. JotDyes are available in 4 colors to suit your research needs (JotDye-430/JotDye-490/JotDye-600/JotDye-800).
Key features: Water soluble, full membrane intercalation, stable binding, no dye exchange, no micelle formation, eliminates false positives, simple workflow, and short incubation time
Selected representative data:

Flow cytometry analysis of EVs labelled with JotDye. Dot plot of (a) unstained EVs, (b) EVs labelled with 1 μM JotDye, (c) 1 μM of JotDye only and (e) 1 μM of commercially available PKH26 dye only.

Uptake of JotDye-Labeled EVs in cell culture. Fluorescence micrograph of HEK-293T cells treated with 20 ug of PC-3 EVs stained with JotDye-600 (red). Nuclei stained with SYTO Far red (blue).

Imaging flow cytometry of cells post-incubated with JotDye-labeled PC3 EVs. JotDye-labeled EVs appeared as green spots. Cells lacking EVs displayed no fluorescence, as free JotDye was eliminated during purification.

Confocal imaging analysis of endogenously stained EVs obtained from JotDye-labeled A549 cells reveals JotDye (green) inside late endosomes (red), indicating their involvement in EV production.
Highlights
EVfect Transfection kit (JOT-EV-T1) – Designed to facilitate the efficient loading of nucleic acid payloads into EVs

EVfect demonstrated high loading efficiencies for both ASOs and plasmid DNA.

EVfect outperformed Exo-fect in gene knockdown efficacy, with Lipofectamine 3000 as the positive control.

EVs loaded with DNA using EVfect showed significantly lower toxicity in cells compared to commercially available reagents.

Mice injected with nanoluciferase-loaded EVs using EVfect displayed continuous and stable luciferase activity at the tumor site over time.
EVcraft Surface modification kit (JOT-EV-SM1) – Designed for the covalent conjugation of any molecules of choice

Approximately 99.8% of the EVs were successfully conjugated with both IgGs.

EVcraft enables the covalent display of up to 700 IgGs on each individual vesicle.

EVcraft enables specific binding of α-CD137-EVs to CD137-expressing T cells.

EVcraft-enabled selective uptake of α-EGFR-EVs by EGFR-positive tumor cells.
